چکیده

In the study of 782 biopsy specimens from 1 95 patients during and after chemotherapy we compared the numbers of Mycobacterium /eprae stained by the Fite-Faraco (FF) and the Gomori methenamine-silver (GMS) techniques. In many patients large amounts of non-acid-fast M. /eprae or remnants thereof remained 66 months after starting effective multidrug therapy. The GMS stain is a useful method for assessing the efficacy of methods for enhancing bacillary clearance in multi bacillary leprosy patients.

In leprosy the presence of mycobacteria in the nerve is an essential factor in the cycle and spectrum of Mycobacterium leprae infection.

A total of 220 untreated leprosy patients who underwent parallel skin and nerve biopsies are included in this study, which is intended to evaluate the extent of previously reported differences in bacillary load between skin and nerve lesions in leprosy and to describe the response of peripheral blood lymphocytes to Mycobacterium leprae antigens in such patients. In 161 patients out of the 220, ...

A RAPID AND SIMPLE METHOD OF DNA EXTRACTION FROM CLINICAL SPECIMENS CONTAINING MYCOBACTERIUM LEPRAE FOR PCR ASSAYS

We describe a rapid, simple, and reliable procedure for routine isolation of Mycobacterium leprae DNA from slit-skin swab specimens. This one-step DNA extraction method is based on the utilization of ChelexR 100, a chelating ion exchange resin. Slit-skin swab specimens from seven untreated leprosy patients at Baba Baghi Leprosy Hospital in Iran were processed by this procedure. The polymer...

THE DETECTION OF Mycobacterium tuberculosis BY PCR IN SPUTUM SAMPLES FROM LONG-TERMTREATED LEPROSY PATIENTS: ASSOCIATIONS WITH SKIN TEST RESULTS AND IMMUNOTHERAPY WITH Mycobacterium vaccae

As part of a series of investigations at Baba Baghi Leprosarium in lran, 44 long-treated leprosy patients were selected for our study. Samples of early morning sputum were obtained from each patient, examined by microscopy for acid-fast bacilli (AFB), and cultured for tubercle bacilli. These tests were negative, but the polymerase chain reaction (PCR) for an insertion sequence believed to b...

Viability of Mycobacterium leprae in skin and peripheral nerves and persistence of nerve destruction in multibacillary patients after 2 years of multidrug therapy.

The pathological changes, bacterial load, and viability of Mycobacterium leprae in the skin and nerves of nine lepromatous leprosy patients who had undergone 2 years of multidrug therapy (MDT) were studied. M. leprae and varying amounts of their remnants were present in the nerves and skin of all but one patient. M. leprae isolated from skin biopsies of six patients and nerve biopsies of nine p...

Presence of M. leprae in tissues in slit skin smear negative multibacillary (MB) patients after WHO-MBR.

This study looked for M. leprae in the lymph node, nerve and skin of multibacillary (MB) leprosy patients who become slit skin smear negative after the completion of WHO-MBR. Twenty-five WHO-MBR-treated multibacillary leprosy patients were studied; borderline lepromatous (BL) leprosy (n = 11) and lepromatous (LL) leprosy (n = 14)). Fifteen patients had reaction (erythema nodosum leprosum 11, up...
